چکیده

S TUDIES OF THE enzyme content of leukocytes reported in the past’9 have been performed on mixed cell suspensions. Some attempts to estimate the enzyme values for individual cell types have been made2, 3. 8 on the basis of complex calculations after multiple assays of mixed cell suspensions with varied cell differential counts. The technic of separation of leukocytes on glass bead 12 now permits a more direct approach to this problem. The present study reports the results of assays of DNA polymerase and of five representative carbohydrate metabolizing enzymes ( glucose-6-phosphate dehydrogenase, 6-phosphogluconate dehydrogenase, lactic dehydrogenase, glyceraldehyde-3-phosphate dehydrogenase and isocitric dehydrogenase) performed on normal and leukemic leukocytes which were separated on glass columns.

Malic and Lactic Dehydrogenase Isozymes of Normal and Leukemic Leukocytes Separated on Glass Bead Columns

IN A RECENT STUDY’ of the enzyme content of normal and leukemic leukocytes separated on glass bead columns, it was shown that meaningful comparisons could not I)e made with mixed cell populations. The work of l)ioguardi et . 2-1I iiidicates that this also holds true for assays of lactic dehydrogenase ( LDH ) isozymes. Comparison, for example, of lymphatic leukemia cells with norinal nlixed leuk...

Deoxycytidine pathway in separated normal and leukemic leukocytes with effects of cell culture.

Enzymes of dC metabolism were studied in glass-column-separated normal and leukemic cells. Supernatants of sonicated cells were incubated with dC-3H or dCMP-3H and the end products separated with paper chromatography. Greatest recovery of dCTP, dUMP, and TTP occurred with myeloblasts and monoblasts. Mature granulocytes, and normal and leukemic lymphocytes gave the greatest breakdown to uracil. ...

Lactate formation and glyoxalase activity in normal and leukemic human leukocytes in vitro.

In their review of the carbohydrate metabolism of leukocytes, Beck and Valentine (4) stated that normal myeloid leukocytes possess a high rate of aerobic glycolysis. Various reports (8, 9, 11) have indicated that, quantitatively, leukocytes from persons with leukemia do not have this capacity to make lactic acid aerobically. However, leukemic leukocytes seem to possess an anaerobic glycolysis, ...

Relationship between RNA-directed DNA polymerase (reverse transcriptase) from human acute leukemic blood cells and primate type-C viruses.

An RNA-directed DNA polymerase was isolated from the peripheral blood leukocytes of a patient with acute myelomonocytic leukemia by successive purification of a particulate cytoplasmic fraction with endogenous, ribonuclease-sensitive DNA polymerase activity. Like RNA-directed DNA polymerase from mammalian type-C virus, the human leukemic cell enzyme efficiently utilized (A)(n).(dT)(12-18) and (...
